Cleavage caspase
Webcleavage: [noun] a fragment (as of a diamond) obtained by splitting. WebCaspase-1 cleaves inflammatory cytokines such as pro-IL-1ß and interferon-γ inducing factor (IL-18) into their mature forms (1,2). Like other caspases, caspase-1 is proteolytically activated from a proenzyme to produce a tetramer of its two active subunits, p20 and p10.

The crystal structure of the post-cleavage complex between p35 and caspase-8 shows that p35 forms a thioester bond with the active site cysteine of the caspase.

WebCaspases are expressed as inactive precursors inside cells and must be activated to cleave specific target substrates. Pro-apoptotic caspases cleave a larger subset of intracellular proteins to mediate cell suicide by apoptosis, while caspases involved in immunity can kill cells by alternative non-apoptotic mechanisms. WebCommitment to this pathway generally involves caspase-mediated protein cleavage and was found to induce cross-presentation of a specific antigen repertoire under certain inflammatory conditions. We aimed to assess the significance of the CD8 T cell population reactive against such caspase-cleaved apoptotic self-antigens in pancreatic islets of ...
